Crohn’s & inflammatory bowel disease
Remission means disease activity is controlled; it does not necessarily mean the disease is gone forever.
Antibody medicineFDA-approved for specified patients
Crohn’s disease & ulcerative colitis: ustekinumab
2+Minimum age in the cited US pediatric indications
Stelara’s Crohn’s indication expanded to children aged two and older with moderately to severely active disease. FDA also reports approval for pediatric ulcerative colitis aged two and older.
The UC decision includes evidence from a 52-week study in 112 children aged 3–17, alongside adult efficacy and pediatric pharmacokinetic and safety evidence. The Crohn’s approval date is 15 April 2026.
What this does not establish
These are treatments for disease control, not proof of a cure. UC safety information includes infections, headache and abdominal pain. Approval is jurisdiction- and indication-specific; individual suitability requires clinical assessment.
No maintenance placebo group; subgroup denominators not supplied in the release.

What was measured or reported
Sponsor-reported maintenance results: Crohn’s endoscopic response was 55% at 900 mg and 41% at 450 mg after 44 maintenance weeks. The extension enrolled 130 initial responders across Crohn’s and ulcerative colitis, after 14 induction weeks. In UC, clinical remission was 58% and 47%, respectively.
What this does not establish
Selected responders were re-randomized between doses; there was no maintenance placebo group. These are not response rates for all starting patients. Crohn’s subgroup denominators are not supplied in the cited release. Endoscopic response means improvement on an intestinal examination, not cure. Reported adverse events included respiratory infections and hypertension. Sponsor report, not an independent comparative verdict.
Stem-cell-derived dopamine-cell precursors were transplanted into the brain to investigate replacement of lost cells. This is cell therapy, not gene editing.

What was measured or reported
The open-label trial used two dose groups of four participants and 12 months of immunosuppression. No serious adverse events were attributed to the cell product; no graft-induced dyskinesias or MRI evidence of tumors were reported during this follow-up.
What this does not establish
One participant died from a pulmonary infection. The paper highlights risks associated with immunosuppression. A small uncontrolled safety trial cannot establish effectiveness, disease reversal or a cure. Follow-up continues to 36 months.
The exPDite-2 registry describes a randomized, double-blind, sham-surgery-controlled phase 3 study. Earlier peer-reviewed phase 1 evidence involved 12 participants, one year of immunosuppression and exploratory improvement in motor measures. A sponsor-funded conference poster reports four-year follow-up of the 12 phase 1 participants at US and Canadian centres. It reports no deaths or discontinuations due to treatment-emergent adverse events, but ten serious adverse events, including a surgery-related seizure. One participant withdrew after the four-year visit.
What this does not establish
The phase 1 study had no control group. Planned enrollment is not the number successfully treated. A later trial phase does not establish efficacy or approval; surgery and immunosuppression remain important burdens. The newer follow-up is a conference report from the same uncontrolled cohort, not independent replication or a new randomized result. It does not establish a cure and adds no daily score credit.
A viral vector delivers instructions for GDNF, a protein intended to support dopamine neurons. It does not replace lost cells or directly edit a Parkinson’s mutation.
The phase 1b study was open-label and uncontrolled. REGENERATE-PD is a randomized, double-blind, surgically controlled phase 2 trial. An April 2026 sponsor update described manufacturing supply for that trial.
What this does not establish
Small early studies cannot establish disease modification. RMAT designation is regulatory support for development, not approval. Definitive benefit and long-term safety remain under study.
An AAV vector delivered instructions for two dopamine-synthesis enzymes, tyrosine hydroxylase and aromatic L-amino acid decarboxylase, to the putamen. This is investigational gene delivery, not an approved treatment or gene editing.
The multicentre, open-label dose-escalation study enrolled ten people with moderate-to-advanced Parkinson’s disease. The 12-month primary safety outcome reported no dose-limiting toxicities or drug-related serious adverse events; 23 reported adverse events were judged unrelated to BBM-P002 and were mainly mild and transient.
What this does not establish
The study had no randomized control group, enrolled only ten participants and was designed primarily to assess safety and tolerability. It does not establish efficacy, durability, disease modification, independent replication or a cure, and adds no daily-v1 score credit.
Gene deliveryPhase 1/2 report; programme terminated early
Parkinson’s: OXB-102 / Axo-Lenti-PD
6Participants treated before sponsor insolvency
A lentiviral vector delivered three genes used in dopamine synthesis to the brain. The study ended early after the sponsor became insolvent; this is not an approval or evidence of a cure.

What was measured or reported
Investigators reported records for six treated participants: two at a lower dose and four at a higher dose. Eight serious adverse events occurred; one was considered definitely related to the intervention. All resolved without reported sequelae. Efficacy data were limited, with possible motor-function stabilization in most participants.
What this does not establish
The open-label, dose-escalation study was small, uncontrolled and prematurely terminated. Possible stabilization cannot establish efficacy, durability, disease modification or a population benefit. The paper does not qualify for daily-v1 event credit.
Gene delivery adds genetic instructions. Gene editing changes DNA. Approved uses remain specific to a disease and eligible population.

Sickle cell disease & beta-thalassemia: Casgevy
8 / 8Evaluable children with sickle cell disease met the specified endpoint
FDA expanded Casgevy to ages two and older for sickle cell disease with recurrent vaso-occlusive crises or transfusion-dependent beta-thalassemia.

What was measured or reported
In children aged 5–11, all 8 efficacy-evaluable SCD participants from an 11-patient study had at least 12 consecutive months without protocol-defined severe crises within 24 months after infusion. In TDT, 8 of 9 evaluable participants from a 15-patient study achieved 12-month transfusion independence. Approval for ages 2–4 involved extrapolation.
What this does not establish
The patient’s blood stem cells are edited outside the body and reinfused after intensive conditioning. Risks include infection-related complications, engraftment problems and unintended edits. Small evaluable groups and limited follow-up do not imply a guaranteed lifetime cure.
FDA added a boxed warning for serious liver injury and acute liver failure after fatal cases in non-ambulatory pediatric patients, and removed the non-ambulatory indication.
What this does not establish
Approval does not mean risk-free or curative. Eligibility and monitoring must follow the current prescribing information. This is a safety milestone, not evidence that Duchenne has been solved.
Replacing insulin-producing cells is a different approach from editing genes.
Cell therapyPhase 1/2 data; phase 1/2/3 programme
Type 1 diabetes: zimislecel
10 / 12Full-dose participants insulin-independent at month 12
Investigational stem-cell-derived islets aim to restore insulin production in people with severe hypoglycemia and impaired awareness. This is cell replacement, not gene therapy.
Ten of 12 full-dose participants with at least one year of follow-up no longer needed external insulin at month 12. The data cutoff was October 2024. Vertex’s August 2026 update reports continued enrollment and dosing in the phase 1/2/3 study.
What this does not establish
This small selected group does not represent everyone with diabetes. Immunosuppression is required. Two deaths were reported in the study programme; investigators did not attribute them to zimislecel itself. The regimen and procedure carry risks, and insulin independence at one year is not proof of a permanent cure.

AI-assisted discovery: ART enzyme systems
EarlyNo demonstrated patient treatment
Anthropic reports that Claude identified defining features of array-associated reverse transcriptases (ART), a system found mainly in viruses that infect bacteria.

What was measured or reported
The team describes a reverse transcriptase, a neighboring protein and repeating DNA sequences. Initial lab work detected short RNAs from the arrays. The system’s underlying enzyme had been identified previously; the reported discovery concerns the wider arrangement and its features.
What this does not establish
Its biological function remains unresolved. CRISPR-like repeats do not establish a working gene-editing tool, safety in humans or a therapy for Crohn’s or Parkinson’s. Independent validation and substantial further work are needed.
Early studies often test safety in small groups. Later controlled trials ask whether benefits exceed those of a comparison treatment or placebo. Trial phase is not a percentage of progress toward a cure. Different endpoints and populations cannot be ranked by response percentages alone.
Endoscopic response means improvement seen during an intestinal examination. Clinical remission means disease activity falls below a specified threshold. Insulin independence and crisis-free periods describe measured intervals, not guaranteed lifelong outcomes. Evaluable participants are the subset with sufficient data for a particular analysis.
